Design Notes

T7 + afp1 part was synthesized as a gBlock from IDT. To remove an internal PstI restriction site, a silent mutation (G-A) was introduced at basepair 331 of the construct. Due to the high GC content of the gene and synthesis limitations, three more silent mutations were introduced to improve gBlock efficiency. These mutations were are located at base pair numbers 115, 133, and 238. The mutation at 115 is C-A and the mutations at base pair numbers 133 and 238 are both C-T.
